This document contains detailed explanation about the methods used for the automated analysis of punctate and diffuse fluorescence in ALM neurites.
We will use the image below as an example to illustrate how the code functions. This image shows a dual color fluorescence micrograph of an ALM neuron, where MEC-4 is labeled in green and NID-1 is labeled in red. Both proteins have punctate localization along the ALM. The raw data for this image was collected on the Keyence BZ-X800 inverted epifluorescence microscope and the overlapping panels were stitched together using the proprietary automated image stitching software from Keyence.
